  To provide culture media and buffers for the investigators who are part of the DERC
   
    In addition to its main objective, the Media Core provides a secondary site of cell storage in liquid nitrogen for DERC investigators, several high volume buffers, and testing for mycoplasma when requested.

Background:
Cultured cells are an indispensable model for studies in all aspects of diabetes and endocrinology, including signal transduction, metabolism, gene expression, protein synthesis and secretion. Investigators at the Joslin Diabetes Center culture a variety of specialty cell types including retinal cells, renal mesangial cells, cardiomyocytes, adipocytes, and islet cells.

In addition, cultured human cells, such as fibroblasts and transformed lymphocytes, are used to study, and define gene polymorphisms that may be related to the development of diabetes and its complications. Because of the multiple uses and many advantages provided by cultured cells, most investigators participating in this DERC are using cultured cells in their studies and have placed a steady increase in demand on Media Core services.


Summary of the Past and Present Use of the Media Core:

The users represent all sections at Joslin Diabetes Center including Immunology, Islet Transplantation and Cell Biology, Metabolism, Cellular and Molecular Physiology, Molecular Biology, Epidemiology and Genetics, Vascular Cell Biology, Obesity, and Glycoproteins and Biomembranes.

A survey was conducted last year to help evaluate the needs and satisfaction of DERC investigators regarding Media Core services. Most users were satisfied with the quality of the media with the exception that some users desired even greater quantities of media and buffers. Thus, we have satisfied the first and the main specific aim of this Core, which is to provide culture media and buffers for the investigators who are part of the DERC. We also have taken on the responsibility of making several molecular biology media and buffers over the grant period. The Media Core has increased its efficiency and productivity in providing media and buffers. In addition, we deliver these media to each laboratory and collect the empty bottles for recycling
